What Is Magnesium Glycinate? Why the Glycine Half Is Not Filler

If you have compared magnesium supplements, you have seen the suffixes: oxide, citrate, glycinate, malate, threonate. They are often presented as flavors of the same thing. They are not. Magnesium is a metal ion. It cannot be put in a capsule by itself; it has to be bound to something, and the something is half the story. In magnesium glycinate, that something is glycine, the smallest amino acid in the body. Understanding why a formulator would choose glycine over a cheaper partner is the clearest way to understand the product on your nightstand.

Magnesium Needs a Partner

In the body, magnesium is the fourth most abundant mineral and a required cofactor for more than 300 enzyme systems, including the ones that make and spend ATP, the cell's energy currency (Institute of Medicine, Dietary Reference Intakes for Calcium, Phosphorus, Magnesium, Vitamin D, and Fluoride, 1997). In a capsule, it exists as a salt or a chelate: a magnesium ion held by a counter-ion or a ligand. Magnesium oxide pairs it with oxygen. Magnesium citrate pairs it with citric acid. Magnesium glycinate pairs each magnesium ion with two glycine molecules, which is why the fully reacted form is properly called magnesium bisglycinate.

The partner matters for three practical reasons. It decides how much of the compound is actually magnesium (the elemental fraction). It decides how the compound dissolves and moves across the gut wall. And it decides what else you are swallowing, because the partner does not vanish; it is absorbed and used too.

What a Chelate Is

The word chelate comes from the Greek for claw. In a chelate, the ligand grips the metal ion at two or more points, forming a ring. Glycine has an amino group and a carboxyl group, and both can bind magnesium, so two glycines close around one magnesium ion like a pair of pincers. The result is a neutral, stable molecule rather than a loosely paired salt.

Why that helps is a matter of chemistry in the small intestine. Free magnesium ions compete with calcium and other minerals for the same transport channels, and they readily form insoluble complexes with phosphates and phytates from food. A chelated magnesium is shielded from some of that. Research on amino acid chelates suggests that part of the dose may be absorbed intact through pathways used for small peptides, in addition to the usual mineral channels, though the size of that contribution in humans is still debated (Schuchardt JP, Hahn A. Current Nutrition & Food Science, 2017;13(4):260–278. PMID 29123461).

What the Research Says About Glycinate Specifically

Direct human trials of magnesium glycinate are fewer than its popularity suggests, and it is worth saying so plainly.

Journal of Parenteral and Enteral Nutrition (1994): Schuette et al. compared magnesium diglycinate with magnesium oxide in patients who had undergone ileal resection, a group with impaired absorption. Using a stable-isotope method, the researchers found the glycinate form was absorbed more completely than the oxide form in these subjects and was well tolerated. The sample was small and the population specific. PMID 7815675

Magnesium Research (2001): Firoz and Graber measured the bioavailability of four commercial magnesium preparations in healthy volunteers. Magnesium oxide showed the lowest fractional absorption, around four percent, while the organic salts tested were meaningfully higher. Glycinate was not among the four, but the study established the gap between oxide and organic forms that most later comparisons rest on. PMID 11794633

Magnesium Research (2005): Coudray et al. compared ten organic and inorganic magnesium salts in magnesium-depleted rats using stable isotopes. Organic forms were generally more bioavailable than inorganic ones. The study is animal work and is cited here for the pattern, not the numbers. PMID 16548135

Taken together, the evidence supports a modest, consistent claim: chelated and organic magnesium forms tend to be absorbed better and tolerated better than magnesium oxide, and glycinate belongs to that group. It does not support a claim that glycinate is uniquely superior to every other organic form.

The Glycine Is Doing Its Own Work

Here is the part most labels skip. A 300 mg elemental dose of magnesium from bisglycinate brings roughly 1.5 to 2 grams of glycine with it. Glycine is an inhibitory neurotransmitter in the spinal cord and brainstem and a co-agonist at NMDA receptors. In small human trials, 3 grams of glycine taken before bed was associated with improved subjective sleep quality and, in one polysomnographic study, a shorter time to reach slow-wave sleep (Bannai M, Kawai N. Journal of Pharmacological Sciences, 2012;118(2):145–148. PMID 22293292). The dose in those trials was higher than what rides along in a magnesium capsule, so the honest framing is that glycine is a partner that fits the evening rather than a second active ingredient at a studied dose.

Compare the alternatives. Oxide brings oxygen, which does nothing. Citrate brings citric acid, which is fine but also draws water into the bowel, the source of its laxative reputation. Glycine brings a calming amino acid. It is this fit between partner and purpose that led us to make bisglycinate two thirds of the magnesium in N–01 Nightly Formula.

Important Context

Magnesium glycinate is not a sedative and does not act like one. The studies above measured absorption and, separately, glycine's effects at doses above what a magnesium capsule carries. Individual absorption varies with magnesium status, kidney function, and what else is in the stomach. Anyone taking medication, particularly antibiotics, bisphosphonates, or diuretics, or anyone with reduced kidney function, should talk to a clinician before adding magnesium in any form.

The Bottom Line

Magnesium glycinate is magnesium held by two glycine molecules. The chelate structure shields the mineral in the gut, the form is consistently better absorbed and tolerated than magnesium oxide in the available research, and the glycine itself is a plausible fit for an evening formula rather than dead weight. The direct trial base for glycinate is thin, and we would rather tell you that than round it up.
